ABSTRACT:

Objectives: To address the clinical evidence that may support the fact that subcutaneous administration of ozone (O3) has anti-hyperalgesic effects probably acting by modifying specific pain targets.

Methods: Fifty-two patients attending the Medinat Clinic, Camerano, Ancona, Italy between October 2004 to October 2009 were eligible to participate in the study. Panoramic photos of painful and not painful areas submitted to oxygen (O2)/O3 treatment
injection were analyzed to detect the intensity of erythema.

Results: The erythematic areas after O3 subcutaneous puncture showed significant (p<0.05) increment (83±5%) in the painful area versus 7±6% in the contralateral area.

Conclusion: The surrounding erythema observed during O3 intervention should explain at least in part, its interaction with some pain mediators. This may involve algesic mediators or receptors. The analgesic mechanism induced by O2/O3 may involve 2
independent steps: a short-term mechanism that may correspond with the direct oxidation on biomolecules, and a long-term mechanism that may involve the activation of antioxidant pathways. Further studies are needed to support the biochemical analgesic mechanism of O3 therapy.

ABSTRACT:

Basic fibroblast growth factor (bFGF) is a pleiotropic mitogen which plays an important role in cell growth, differentiation, migration and survival in different cells and organ systems. Application of bFGF has been shown to promote cellular proliferation and collagen synthesis in vivo. FGF is markedly up-regulated following bone or tendon injury and active at multiple stages of the healing stimulation process, local blood circulation, lipolysis and smooth muscle. Looking at the physical and chemical properties of the ozone molecule, the present work deals with its possible therapeutic action as an FGF activator. Incubation (2 h) of platelet-rich plasma with 80 μg/mL O2/O3 increases the basal concentration of FGF by approximately 600%. This fact in combination with previous demonstration of the stimulating action of O3, releasing other platelet factors may potentially allow autologous treatment in aesthetic and clinical tissues conditions in which FGF has a leading role. The versatility and broad beneficial effect of ozone has become evident in orthopedics, cutaneous and mucosal infections as well as in dentistry. The induction of FGF and other growth factors by ozone can support and potentiate those applications.

ABSTRACT:

Ozone therapy is widely used in many countries since many years. Oxidative preconditioning is the essential bases of ozone therapy. We show data collected on patients treated in the last three years for disorders related to pain either in sport traumatism (232 subjects) or  derived for inflammatory disorders (770 subjects). The evolution of patients was follow using the Overall Patient Satisfaction Scale. The maximal score (8-10), corresponding qualitatively to “ very good” was reached in 80% of patients. A rapid and sustaining success was reached in pubalgy to a score  8-10 as soon as the theraoy was started. In patients with pain from inflammatory disorders a progressive evolution in time was reached to a score 8-10. However, the maximal score in joint inflammatory diseases was 6-8(qualitatively “good”). No side effects were recorderd at short and long-term follow-up. In our experience, O2/O3 treatment of pain and inflammatory diseases has revolutionized the approach to radiculopathy and articular disease pains manage.

The therapeutic use of ozone (O2-O3) gas is controversial. Some authors claim the gas mixture is toxic and therefore out of the question, whereas, others hold that it is useful and effective when used following a certain method to treat degenerative diseases, such as knee osteoarthritis, for which the gas has been used empirically. The present work studied the effects of contact of O2-O3 with healthy knee tissues of Wistar rats and compared them to intra-articular injection of a substance known to damage the cartilage.

 Thirty-six Wistar rats were studied in two groups. Mono-iodoacetic acid (MIA) was injected in the first group in a single dose και, whereas O2-O3was injected in the second group in frequent doses three times a week for three weeks. The rats were killed 40 days later and articular cartilage and surrounding tissues were studied histologically. MIA caused degenerative osteoarthritis gradually deteriorating at the knees of the first group whereas no major changes were observed in those of the second group. We conclude that following the methodology of our study medical ozone appears to be safe for use.

The aim of this review was twofold. The first was to correlate the empirically determined ozone dose to the presence and amount of antioxidant capacities present in several body fluids and tissues. Although some data are uncertain, we have reached the conclusion that ozone must be administered with care and the doses, so far assessed by trial and error, are reasonable and apparently useful. What is most surprising is that a theoretically offensive stimulus, represented by a sudden administration of oxygen-ozone, is able to reactivate natural defense mechanisms in a pleiotropic fashion restoring health and eliminating pain…
